Seit der Antike werden Naturstoffe in großem Umfang zur Behandlung von Krankheiten eingesetzt und sie spielen nach wie vor eine wichtige Rolle in der Arzneimittelforschung. Ziel dieser Arbeit war daher die Identifizierung und Charakterisierung neuer Naturstoffe aus Cyan obakterien für die Arzneimittelentwicklung. Um dieses Ziel zu erreichen, wurden drei verschiedene Ansätze verfolgt. Ein strukturgeleitetes Screening wurde mit HaloSeeker vorgenommen, um neue halogenierte Naturstoffe zu identifizieren. Darüber hinaus wurden zwei verschiedene Bioassay basierte Screening Kampagnen durchgeführt, um Immunmodulatoren und Quorum Sensing Inhibitoren zu identifizieren. Alle drei Screening Ansätze waren erfolgreich bei der Identifizierung neuer bioaktiver Verbindungen aus Cyanobakter ien und verdeutlichen, dass Cyanobakterien eine ergiebige Quelle für bioaktive Naturstoffe sind.
Since ancient times natural products have been widely used to treat diseases, and they continue to play an important role in drug discovery. Thus, this thesis aimed to identify and characterize new natural produ cts from cyanobacteria for drug development purposes. Three different approaches were used to achieve this goal. A chemistry guided screening was performed with HaloSeeker in an effort to identify novel halogenated natural products. In addition, two distin ct bioassay based screening campaigns were conducted to identify immunomodulators as well as quorum sensing inhibitors. All three screening approaches were successful in identifying new bioactive compounds from cyanobacteria and illustrate that cyanobacter ia are a prolific source for bioactive natural products.
