  • Beschreibung: <jats:sec> <jats:title>Background</jats:title> <jats:p>The association of type 2 diabetes mellitus (T2DM) with the <jats:italic>KCNJ11, CDKAL1, SLC30A8, CDKN2B,</jats:italic> and<jats:italic> FTO</jats:italic> genes in the Russian population has not been well studied. In this study, we analysed the population frequencies of polymorphic markers of these genes.</jats:p> </jats:sec> <jats:sec> <jats:title>Methods</jats:title> <jats:p>The study included 862 patients with T2DM and 443 control subjects of Russian origin. All subjects were genotyped for 10 single nucleotide polymorphisms (SNPs) of the genes using real-time PCR (TaqMan assays). HOMA-IR and HOMA-<jats:italic>β</jats:italic> were used to measure insulin resistance and <jats:italic>β</jats:italic>-cell secretory function, respectively.</jats:p> </jats:sec> <jats:sec> <jats:title>Results</jats:title> <jats:p>The analysis of the frequency distribution of polymorphic markers for genes <jats:italic>KCNJ11, CDKAL1, SLC30A8</jats:italic> and <jats:italic>CDKN2B</jats:italic> showed statistically significant associations with T2DM in the Russian population. The association between the <jats:italic>FTO</jats:italic> gene and T2DM was not statistically significant. The polymorphic markers <jats:italic><jats:ext-link xmlns:xlink="http://www.w3.org/1999/xlink" ext-link-type="uri" xlink:href="http://www.ncbi.nlm.nih.gov/snp/?term=rs5219">rs5219</jats:ext-link></jats:italic> of the <jats:italic>KCNJ11</jats:italic> gene, <jats:italic><jats:ext-link xmlns:xlink="http://www.w3.org/1999/xlink" ext-link-type="uri" xlink:href="http://www.ncbi.nlm.nih.gov/snp/?term=rs13266634">rs13266634</jats:ext-link></jats:italic> of the <jats:italic>SLC30A8</jats:italic> gene, <jats:italic><jats:ext-link xmlns:xlink="http://www.w3.org/1999/xlink" ext-link-type="uri" xlink:href="http://www.ncbi.nlm.nih.gov/snp/?term=rs10811661">rs10811661</jats:ext-link></jats:italic> of the <jats:italic>CDKN2B</jats:italic> gene and <jats:italic><jats:ext-link xmlns:xlink="http://www.w3.org/1999/xlink" ext-link-type="uri" xlink:href="http://www.ncbi.nlm.nih.gov/snp/?term=rs9465871">rs9465871</jats:ext-link></jats:italic>, <jats:italic><jats:ext-link xmlns:xlink="http://www.w3.org/1999/xlink" ext-link-type="uri" xlink:href="http://www.ncbi.nlm.nih.gov/snp/?term=rs7756992">rs7756992</jats:ext-link></jats:italic> and <jats:italic><jats:ext-link xmlns:xlink="http://www.w3.org/1999/xlink" ext-link-type="uri" xlink:href="http://www.ncbi.nlm.nih.gov/snp/?term=rs10946398">rs10946398</jats:ext-link></jats:italic> of the <jats:italic>CDKAL1</jats:italic> gene showed a significant association with impaired glucose metabolism or impaired <jats:italic>β</jats:italic>-cell function.</jats:p> </jats:sec> <jats:sec> <jats:title>Conclusion</jats:title> <jats:p>In the Russian population, genes, which affect insulin synthesis and secretion in the <jats:italic>β</jats:italic>-cells of the pancreas, play a central role in the development of T2DM.</jats:p> </jats:sec>
