    The Gram-positive bacterium Streptococcus pneumoniae is a human-specific pathogen of the upper respiratory tract. The opportunistic pathogen can infect multiple organs and penetrate deeper into the body, leading to local inflammation such as sinusitis and otitis media. Life-threatening infections such as pneumonia, meningitis or sepsis are possible. S. pneumoniae is poorly understood on the metabolome level. Therefore, the aim of this thesis was a comprehensive characterization of the metabolome of S. pneumoniae. Within the present thesis, gas chromatography (GC) and liquid chromatography (LC) coupled with mass spectrometry (MS) as well as nuclear magnetic resonance spectroscopy (NMR) were used to analyze the metabolome of S. pneumoniae, since there are several bioanalytical approaches are necessary to investigate the metabolome and its chemically different metabolites. Within this context Article I summarizes the known metabolome studies of S. pneumoniae. The basis metabolome of S. pneumoniae was elucidated using the aforementioned techniques. Here especially the reproducible growth during the cultivation and the sampling procedure must be developed and optimized. A sampling protocol for the endometabolome of S. pneumoniae was established in this study (Article II). Using this optimized protocol, a comprehensive metabolome analysis was performed in the chemically defined medium.
