In der vorliegenden Arbeit wurde die grundsätzliche Realisierbarkeit eines enzymatischen in vitro Produktionsprozesses für 1,3-Propandiol (1,3-PDO) aus Glycerol demonstriert. Dabei konnte eine sehr hohe Ausbeute von ca. 0,95 mol 1,3-PDO/mol Glycerol erzielt werden. Der klar identifizierte produktivitäts- und kostenbestimmende Faktor stellt in dem in vitro System die Glyceroldehydratase mit ihrem mechanismausbasieren Suizideffekt dar.
In the present work the feasibility of engineering an enzymatic in vitro production process for 1,3-propanediol (1,3-PDO) from glycerol was demonstrated. In this novel bioprocess, a very high production yield of about 0.95 mol 1,3-PDO/mol glycerol was reached. The bottleneck of the process regarding costs and productivity is considered to be mainly related to the inactivation of glycerol dehydratase through its mechanism-based suicide-effect.
