Description: Methylation of the lithio salt of 2‐methylthio‐2‐thiazoline with methyl iodide 14C or methyliodide D3 followed by thiazoline displacement with benzyl bromide has given rise to ethyl bromide ‐2‐14C and ethyl bromide‐2‐D3. The Grignard reagents from the latter, treated with 2,2‐diphenyl‐4‐dimethylaminovaleronitrile gave methadone‐1‐14C and methadone‐1‐D3, purified by silicagel adsorption column chromatography. Overall yields = 14–20% based on labelled ICH3, methadone‐1‐14C (SA = 36 mci/mM), methadone ‐1‐D3 (99% isotopic enrichment).
