<jats:p>
To increase the frequency of homologous recombination, we inactivated the
<jats:italic>KU80</jats:italic>
homologue in
<jats:italic>Aspergillus fumigatus</jats:italic>
(named
<jats:italic>akuB</jats:italic>
<jats:sup>KU80</jats:sup>
). Homologous integration reached about 80% for both calcineurin A (
<jats:italic>calA</jats:italic>
) and polyketide synthase
<jats:italic>pksP (alb1</jats:italic>
) genes in the
<jats:italic>akuB</jats:italic>
<jats:sup>KU80</jats:sup>
mutant to 3 and 5%, respectively, when using a wild-type
<jats:italic>A. fumigatus</jats:italic>
strain. Deletion of
<jats:italic>akuB</jats:italic>
<jats:sup>KU80</jats:sup>
had no influence on pathogenicity in a low-dose murine infection model.
