The detachment of podocytes from the glomerular basement membrane leads to glomerulopathies. Loss can be induced by increased mechanical stress such as hypertension or by a failed interaction of the actin cytoskeleton, focal adhesions and the basement membrane. Research into the actin cytoskeleton and the focal adhesions therefore promises to provide insights into the mechanisms of renal failure. Fluorescence microscopy shows detailed images of these structures. However, subsequent description and quantification is difficult due to unordered distributed arrangements and structures of the filaments and focal adhesion. The aim of this thesis is the development and testing of automated image analysis methods for feature extraction of focal adhesions, the actin cytoskeleton and the cell shape of podocytes.
