Over the last decade, several populations of B cells with regulatory functions have been identified in an increasing number of pathological conditions. Since for several years we have been interested in a population of kidney transplant patients tolerant to their graft, we wanted to explore this regulatory process in these patients. After having shown that tolerant patients had a transcriptional profile with a strong B lymphocyte imprint, we showed that they had a higher number of B cells expressing granzyme B capable of inhibiting the proliferation of effector T cells by a mechanism depending on the contact of the B lymphocyte with its target and on granzyme B. We characterized these B lymphocytes and were able to establish a protocol for the expansion of these cells allowing us to consider their future use in cell therapy.
Au cours de la dernière décennie, plusieurs populations de lymphocytes B avec des fonctions régulatrices ont été identifiées dans un nombre croissant de conditions pathologiques. Alors que depuis plusieurs années nous nous intéressons à une population de patients transplantés tolérants leur greffon rénal, nous avons voulu explorer ce processus de régulation chez ces patients. Après avoir montré que les patients tolérants présentaient un profil transcriptionel avec une forte empreinte lymphocytaire B, nous avons montré qu’ils présentaient un nombre plus élevé de cellules B exprimant le granzyme B capables d''inhiber la prolifération des cellules T effectrices par un mécanisme dépendant du contact du lymphocyte B avec sa cible et du granzyme B. Nous avons caractérisé ces lymphocytes B et avons pu établir un protocole d’expansion de ces cellules permettant d’envisager leur utilisation future en thérapie cellulaire.
